Researchers link 2 biological risk factors for schizophrenia

Johns Hopkins researchers say they have discovered a cause-and-effect relationship between two well-established biological risk factors for schizophrenia previously believed to be independent of one another.

The findings could eventually lead researchers to develop better drugs to treat the cognitive dysfunction associated with schizophrenia and possibly other mental illnesses.

Researchers have long studied the role played in the brain's neurons by the Disrupted-in-Schizophrenia 1 (DISC1) gene, a mutation with one of the strongest links to an increased risk of developing the debilitating psychiatric illness.

In a study published in the journal Molecular Psychiatry, the laboratory of Mikhail V. Pletnikov, M.D., Ph.D., in collaboration with the laboratory of Solomon H. Snyder, M.D., D.Sc., instead looked at the role the DISC1 gene plays in glia cells known as astrocytes, a kind of support cell in the brain that helps neurons communicate with one another.

"Abnormalities in glia cells could be as important as abnormalities in neuronal cells themselves," says Pletnikov, an associate professor of psychiatry and behavioral sciences at the Johns Hopkins University School of Medicine, and the study's leader. "Most gene work has been done with neurons. But we also need to understand a lot more about the role that genetic mutations in glia cells play because neuron-glia interaction appears crucial in ensuring the brain operates normally."

Besides the paranoia and hallucinations that characterize the disease, schizophrenics have cognitive deficits, leaving them unable to think clearly or organize their thoughts and behavior.

Previous studies found that one of the roles of astrocytes is to secrete the neurotransmitter D-serine, which helps promote the transmission of glutamate in the brain, believed to be a key to cognitive function. Schizophrenics have decreased glutamate transmission. It appears, Pletnikov says, that people with DISC1 mutations associated with the psychiatric illness are faster to metabolize D-serine, which leads to a decrease in the apparently crucial transmitter.

In clinical trials, other researchers are trying to boost D-serine levels in people with schizophrenia to see if they can boost cognitive function.

In the new study, the Johns Hopkins researchers found that DISC1 is directly involved in regulating the production of D-serine by the enzyme known as serine racemase.

The researchers found that DISC1 normally binds to serine racemase and stabilizes it. The mutant DISC1 in patients with schizophrenia cannot bind with serine racemase, and instead destabilizes and destroys it. The result is a deficiency of D-serine.

The Hopkins researchers bred mice with the mutant DISC1 protein expressed only in astrocytes and, as predicted, the animals had decreased levels of D-serine. These mice also showed abnormal behavior "consistent with schizophrenia," Pletnikov says. For example, the rodents showed sensitivity to psycho-stimulants that target glutamate transmission. By treating the mice with D-serine, the scientists were able to ameliorate the schizophrenic-like symptoms. Mice without the DISC1 mutation in astrocytes had normal D-serine levels.

Pletnikov says that in the future, researchers hope that they can target the unstable junction between the abnormal DISC1 and serine racemase. If drugs, for example, can be found to increase glutamate transmission in humans, doctors may be able to improve cognitive function in schizophrenics. He says a DISC1 mutation may also be an important risk factor in other psychiatric disorders.

"Abnormal glutamate transmission is believed to be present in patients with bipolar disorder, major depression and possibly anxiety disorders, so our findings could apply to other psychiatric diseases," he says.

There is a long held belief that the stock market is the fore-teller of all that will happen. Somehow it is believed that the market can see into the future, and the rises or falls in the market foretell what will happen with some accuracy. Believers, which includes most people, ignore the dramatic rises in the market before the meltdown of the money market in 2007, or the great crashes of 2008 and 2009. Still, in spite of much evidence to the contrary, this belief persists.

On the other hand, great investors such as Warren Buffet, believe that the market swings irrationally and that you buy when stocks are irrationally depressed.

The Efficient Market Theory
This theory, known as ?The Efficient Market Theory?, is also the basis of Investment Advisors advocating a ?Buy and Hold? approach to investing. The belief is that in due course, the market will always value a stock fairly, and that stocks tend to rise over time. This theory says that all stocks are valued properly because all information necessary to value a stock is available.

A Decision by a Group is Better than an Individual?s Decision
The belief is that the stock market, by being valued by the sum of each individual investment decision, is the total of all of these individual decisions, and therefore is almost always correct. This theory has been generally accepted as for some odd and vaguely understood process, a decision made by discussion among a group, is more often then not, a better decision than would have been made by any individual. It seems that even if the average knowledge, or IQ, or technical knowledge, of the group seems lacking or less than adequate, the decision made by the collective wisdom of this group, is often more accurate and a better decision than a decision made on the subject than any individual, no matter the intelligence or understanding of that individual.

The Market Has All Information on Which to Make a Decision
The same theory, which is that a group understands any situation far better than any individual, states that The Efficient Market Theory means that the stock market has all of the information necessary to make an informed decision. Therefore, somehow all knowledge about every stock is known to the market in general, and the value of that stock, and of all stocks, is established because the market has all information necessary to come to that valuation.

The Efficient Market Theory is Nonsense
The very fact that Facebook could trade at $38 at opening, then over $40, then at $28, and now slightly over $30 is proof that the efficient market theory is nonsense. Another example is often stated by Warren Buffet, who in reference to the idiosyncrasies of the market, asked why General Motors was worth over $20 one year, and under $2 the next year. He remarked that it was the same company, in the same business, doing the same thing, yet depending on when you look at the stock price, it varies dramatically.

Another Warren Buffet quote is ?Most people get interested in stocks when everyone else is. The time to get interested is when no one else is. You can?t buy what is popular and do well.? In other words, the value of any stock fluctuate dramatically. If the stock market was an efficient barometer of value, the price would reflect true value at all times, yet it doesn?t.

Other Indications of the Folly of this Theory
As time passes, more and more of the stock market volume is as a result of program trading. Computers now account for over 80% of the trading volume. These computers trade based on market trends. They rely on sophisticated software that determines whether a stock is poised to rise or fall. What is more interesting, is that because there is so much trading based on computer programs, that stock market rules have had to been continually revised to prevent meltdowns. Computer trading, also know as program trading, call be a self-fulfilling prophecy. As one program trades down, other programs watching, see a lower trade and also trade down to catch the trend. The result can be a precipitous drop in a matter of minutes. So much for fair valuation of stock values.

The point here is that doom and gloom pervades the market, and fear is inhibiting most investors. If a stock is supported by a company with good value, with good underlying assets, and there is no logical reason for it to have fallen so much, then that stock is probably a good value to buy right now. If you don?t buy it now, you will look back in a a few years and bemoan the fact that you didn?t buy when the price was so low. Now is the time to lay the groundwork of getting wealthy.

Now, for the first time since the museum opened in 1929, the public gets to see it as its architects intended. The Rodin Museum reopened Friday after a more than three-year, $9 million renovation that returned all its sculptures to their original locations inside and out, refurbished almost all of them ? only "The Burghers of Calais" has yet to be cleaned up ? and restored the grounds' formal French garden, fountain and reflecting pool.

"It was long overdue," Timothy Rub, director and chief executive officer of the Philadelphia Museum of Art, which runs the Rodin, said at a preview event Thursday. "We have a beautiful site and building and a great collection that had, frankly, lost some of its luster."

The inside galleries were rearranged to emphasize the way many figures in "The Gates of Hell" ? Rodin's colossal masterwork that dominates the museum entrance ? inspired his later iconic sculptures from "The Kiss" to "The Thinker." Behind the scenes, a new air-conditioning system means a swelter-free visit for summer tourists for the first time in decades.

Located between the Philadelphia Museum of Art and the new Barnes Foundation on the Benjamin Franklin Parkway, the classical Beaux-Arts building contains the largest collection of Auguste Rodin's sculptures outside of the Musee Rodin's collections in Paris and Meudon. It was a gift to the city from movie theater magnate Jules Mastbaum, who was introduced to Rodin's work during a 1923 trip to Paris.

Mastbaum hired two France-born architects living in Philadelphia, architect Paul Cret and landscape designer Jacques Greber, to create the limestone museum. Its holdings include more than 140 bronze, marble and plaster sculptures, plus drawings, prints, letters and books.

"Everything you see here, the interior in particular, is a true restoration of one of the most sophisticated buildings ever," curator Joseph Rischl said. "This is as sophisticated as a Parisian dress of 1929."

The Rodin Museum also houses a few works that were created by other artists.

Two small bronze heads, attributed to Rodin when Mastbaum purchased them, were later found to be the work of the artist's collaborator, muse and lover Camille Claudel. In addition, the Musee Rodin permitted Mastbaum to commission a copy of "The Kiss," one of Rodin's best-known works. The marble reproduction in Philadelphia was completed in 1929, 12 years after Rodin's death, by Jacques Greber's father, sculptor Henri Greber.

As is the case in museums around the world, the majority of the Rodin sculptures in Philadelphia's museum weren't cast in bronze until years after his death in 1917.
